Description :
CD4, also known as T-cell surface antigen T4, Leu-3, L3T4, and W3/25, is a single-pass type I transmembrane glycoprotein of the immunoglobulin superfamily (IgSF). CD4 contains 3 Ig- like C2-type (IgC2) domains and 1 Ig-like V-type (IgV) domain in its extracellular region. CD4 is expressed predominantly on thymocytes and a subset of mature T lymphocytes. CD4 serves as a standard marker for the identification of T cell populations. CD4 is expressed along with CD8 on T cells during their early development in the thymus. CD4 or CD8 expression is then lost, giving rise to single positive CD4+ or CD8+ mature T cells. CD4+ cells, also known as T helper (Th) cells, further differentiate into multiple subsets including Th1, Th2, Th17, Tfh, and Treg cells, which are crucial in the regulation of humoral and cellular immunity. In human, CD4 is also expressed on macrophages, neutrophils, monocytes, NK cells, dendritic cells and glial cells in the brain. CD4 is a co-receptor that assists the T cell receptor (TCR) to activate T cells following the interaction with MHC class II molecules on antigen presenting cells (APC). CD4 also functions as a chemotactic receptor for IL­16. In human, CD4 also acts as a co-receptor for the gp120 surface glycoprotein of HIV­1. CD4 is a primary receptor by HIV-1 to gain entry into host T cells.
