Description :
TACI (transmembrane activator and CAML­interactor), also known as CD267, is a single-pass, type III transmembrane glycoprotein in the TNF receptor superfamily (TNFRSF13B). The TNFRSF members have crucial roles in both innate and adaptive immunity as well as in apoptosis. The extracellular region of TACI contains two TNFR-cysteine rich repeats that mediate interactions with its TNF family ligands, including APRIL/TNFSF13 and BAFF/TNFSF13B. TACI binds both ligands with similar high affinity. Within the TNFRSF, it shares the highest homology with BCMA/TNFRSF17. TACI and BCMA can bind both APRIL and BAFF. TACI is expressed on the cell surface of B cells and activated T cells, but not on resting T cells. TACI may play an important role in B cell development, function and regulation. TACI interacts with calcium-modulator and cyclophilin ligand (CAML). TACI mediates calcineurin-dependent activation of NF-AT, NF-κB and AP-1. TACI is also involved in the stimulation of B- and T-cell function and the regulation of humoral immunity. Mutations in the TACI gene are associated with common variable immunodeficiency 2 (CVID2) and immunoglobulin A deficiency 2 (IGAD2) in humans.

The recombinant human TACI-Fc fusion is expressed as a 391 amino acid protein consisting of Ser2 – Ser165 region of TACI (UniProt accession #O14836) and a C-terminal Fc from human IgG1, which exists as a dimer under non-reducing conditions (see the gel image inserted above).
