Description :
DR3 (death receptor 3), also known as AIR, LARD, WSL­1, APO3, TRAMP and TR3, is a single pass type I transmembrane glycoprotein in the TNF receptor superfamily (TNFRSF25). DR3 contains an extracellular region with 4 TNFR cysteine­rich repeats and a cytoplasmic region with 1 death domain (DD) that is also found in other 5 TNFRSF members. DR3 is most closely related to TNFR1 and FAS/CD95, sharing 29% and 23% amino acid sequence identity, respectively. DR3 is abundantly expressed in thymocytes and lymphocytes and detected in lymphocyte-rich tissues such as thymus, colon, intestine, and spleen. It is also found in the prostate. Naïve B and T cells express multiple truncated DR3 isoforms that lack the transmembrane domain. Upon T cell activation, expression of the transmembrane isoform 1 of DR3 predominates. DR3 interacts directly with the adapter protein TRADD to mediate activation of NF-κB and induce apoptosis. DR3 may play a role in regulating lymphocyte homeostasis. Knockout studies in mice suggested the role of this gene in the removal of self-reactive T cells in the thymus. TL1A/TNFSF15, a TNF superfamily ligand, has been shown to bind and activate DR3. On T cells, TL1A induces a costimulatory signal to increase IL­2 responsiveness and the secretion of proinflammatory cytokines. TL1A has also been shown to induce caspase activity and apoptosis.
