Description :
CD360, also known as IL21R (Interleukin-21 receptor), is a single pass, type I transmembrane glycoprotein of the type I cytokine receptor family, type 4 subfamily. CD360/IL21R consists of an extracellular region with 2 fibronectin type III (FN3) domains and a juxtamembrane WSXWS motif, a transmembrane domain, and a cytoplasmic tail with a box 1 motif. The WSXWS motif is necessary for proper protein folding and efficient intracellular transport and cell-surface receptor binding, while the box 1 motif is required for JAK interaction and activation. CD360/IL21R acts as a receptor for IL21 and forms a heterodimeric receptor complex with the common gamma-chain, interleukin 2 receptor gamma (IL2RG), a receptor subunit also shared by the receptors for interleukin 2, 4, 7, 9, and 15. CD360/IL21R transduces the growth promoting signal of IL21, and is important for the proliferation and differentiation of T cells, B cells, and natural killer (NK) cells. IL21 binding leads to the activation of multiple downstream signaling molecules, including JAK1, JAK3, STAT1, and STAT3. Knockout studies suggest a role for the IL21R/CD360 gene in regulating immunoglobulin production. The mutations affecting the IL21R/CD360 gene is associated with IL21R immunodeficiency (IL21RID), an autosomal recessive primary immunodeficiency characterized by B- and T-cell defects and variable dysfunction of NK cells. Patients have normal numbers of lymphocytes, but show defective class-switched B-cells, low IgG, defective antibody response, and defective T-cell responses to certain antigens.
